Description
Synaptosomal‑associated protein 8, or the SNAP‑8 peptide, is a synthetic octapeptide analog of the N‑terminal end of the SNAP‑25 peptide. The peptide is made of eight amino acids and is acetylated at the N‑terminus and amidated at the C‑terminus, resulting in the structure Ac‑Glu‑Glu‑Met‑Gln‑Arg‑Arg‑Ala‑Asp‑NH₂.
Overview
SNAP‑8 was developed to compete with SNAP‑25 protein interactions and bind with vesicle‑associated membrane proteins. SNAP‑25 is a key component in neurotransmitter release, normally interacting with membrane proteins like Synaptotagmin 1 to facilitate fusion of vesicles with the cell membrane and release of neurotransmitter contents.
Molecular docking and experimental models suggest SNAP‑8 may bind to parts of the Synaptotagmin 1 protein responsible for calcium response, potentially destabilizing the SNARE complex that is essential for vesicle fusion. This destabilization could interfere with neurotransmitter release, particularly acetylcholine — a neurotransmitter involved in muscle contraction.
By reducing acetylcholine‑mediated muscle activity, SNAP‑8 is studied in experimental settings for its potential to reduce muscle contractions associated with expression lines and wrinkles in skin models.
